Despite their abundance in nature and the well-established relevance of these alkaloids, their photochemical and photobiological properties as well as their main biological role remain unclear. Moreover, in most of the cases, the fundamental aspects and molecular basis of the mechanisms involved still remain poorly understood. In this review, we summarize the most relevant spectroscopic, chemical and photochemical properties observed in aqueous environment for a representative set of full-aromatic, dihydro- and tetrahydro-βCs. All the fundamental aspects herein discussed provide key insights to theorize and/or surmise about the biological role that these compounds might have.

β-carbolines (βCs) comprise a large variety of naturally occurring alkaloids structurally related to 9Hpyrido[ 3,4-b]indole or norharmane, widely spread in a vast range of phylogenetically distant species, including phototrophic organisms. All the fundamental aspects herein discussed provide key insights to theorize and/or surmise about the biological role that these compounds might have.</dc:description>

The biological and medical importance of photosensitized reactions is mainly related to their participation in processes involved in the development of skin cancer and in photodynamic treatments against cancer and infections. Antioxidant And Anti-Inflammatory Effects Of Anthocyanins In In Vitro, In Vivo, And Human Clinical Trials. Natural Sources And Molecular Pathways Involved
Anales de la Asociación Química Argentina; vol. 112, no. 2
This paper discusses the results of a meta-analysis we recently conducted on eleven human chondrocyte studies, nineteen clinical trials in patients with osteoarthritis, and twenty-two trials in overweight/obese patients. The interaction between osteoarthritis inflammation and obesity, and the subsequent regulation/immunomodulation, was mediated by dietary anthocyanins. The best results were obtained with strawberries, cherries, berries, pomegranates, tropical fruits, rosehips, purple rice, purple corn, kidney beans, and black soybeans, along with the aglycones cyanidin, delphinidin, malvidin, and peonidin, some 3-O-glucosides, metabolites, and acylated anthocyanins from a dark purple-fleshed potato cultivar. Anthocyanins alleviated pain and stiffness in osteoarthritis, contributing to improve daily activity and biometric data in patients, thus highlighting the clinical relevance of dietary anthocyanins for the prevention and management of osteoarthritis and chronic comorbidities. The signaling pathways involved were analyzed at the molecular level.

A photosensitized oxidative crosslinking of proteins (POCP) reaction was applied in air-saturated phosphate buffer solutions of bovine serum albumin (BSA) to obtain soluble protein nanoparticles of approximately 100 nm in diameter. A royal blue LED was used as the excitation source for the photosensitizer molecule ruthenium (II) 2− tris(2,2′-bipyridyl) dication (Ru(bpy)2+ 3 ), in the presence of the electron acceptor persulfate anion (S2O8 ). The redox quenching products prompted the formation of side-chain tyrosyl radicals, which served as intermediaries in the covalent attachment between proteins, leading to the formation of dityrosine (Tyr2) links. However, the dissolved oxygen competes efficiently with S2O2− 8 to quench the excited photosensitizer, thereby generating singlet molecular oxygen (1O2), which reacts with electron-rich protein residues, which in turn induce an additional oxidative pattern of BSA. Consequently, under air-saturated conditions, the POCP gives rise to a series of oxygen-dependent and -independent reactions, resulting in the protein crosslinking with oxidative modifi cations. The esterase-like activity efficacy of BSA oxidized solely by 1O2 and after the formation of oligomeric protein nanoparticles by POCP was reduced by 51 % and 73 %, respectively, as compared with that of the native BSA. The combination of the oxidative degradation of key residues in the active sites and steric impediment due to protein oligomerization was found to be associated with this result.

3,3′-Dityrosine, resulting from the formation of a carbon-carbon bond between two tyrosines (Tyr), is one of the most important modifications of the oxidatively generated damage to proteins. Its formation can induce structural changes in proteins, leading to the loss of their biological function. Interestingly, under UVA radiation, dityrosine can also act as an intrinsic photosensitizer that generates reactive oxygen species and induces chemical modifications in amino acids. Despite the biomedical importance of dityrosine, the information regarding its photosensitized generation and photochemical properties is limited due to the drawbacks inherent to its synthesis.
In this work we studied the photosensitized formation of dityrosine, in the absence and presence of tryptophan (Trp), using pterin (Ptr) as endogenous type I sensitizer. Free Tyr and Tyr incorporated into specially designed peptides were used as target molecules. Our results indicate that the efficiency of dimerization is not affected by the presence of the peptide bond. Nonetheless, incorporation of dityrosine into a peptide chain appears to confer a certain degree of protection, making it more resistant to photosensitized degradation. Moreover, we demonstrated that, within a peptide chain, an adjacent Trp residue promotes the photoinduced dimerization of Tyr, likely due to electron transfer from the Tyr residue to the oxidized Trp. This effect is not observed when the two residues are more distant. Additionally, the presence of free Trp or Trp within the same peptide chain enhances the efficiency of the photosensitized degradation of the free and peptide dityrosine, respectively.

Superparamagnetic iron oxide nanoparticles are used in MRI as T2 or T2* contrast agents and, in combination with relaxometry, enable quantitative analysis of physiological and pathological processes. However, the induced changes in relaxation times are influenced by a complex interplay of the contrast agents’ physicochemical properties. Here, an open-source Monte Carlo simulation pipeline was implemented, which enables the char acterization of these relaxation time changes caused by MRI contrast agents. The simulation tool was validated by showing that simulated relaxation times for iron oxide particles matched the solutions of analytical models of the respective diffusion regimes. For comparison with relaxometry measurements, T2 and T2* of four MRI contrast agents Ferucarbotran, FeraSpin XL, magnetite nanohexagons (MNH@OH) and magnetite nanocubes (MNC@OH) were simulated, using three approaches for modeling contrast agent size and composition: 1) uniform particle sizes using the median hydrodynamic radii; 2) distributed radii corresponding to measured hydrodynamic radius distributions; 3) size-distributed magnetite cores in a coating layer of uniform radius. The simulation accurately reproduced measured relaxation times when appropriate modeling strategies for contrast agent size and composition were used. For FeraSpin XL and MNH@OH, using uniform radii provided good estimates of relaxation times, which was further improved by using the size distributions. For MNC@OH, discrepancies in simulated and measured T2 for all approaches were attributed to particle aggregation. <dc:description>Superparamagnetic iron oxide nanoparticles are used in MRI as T2 or T2* contrast agents and, in combination with relaxometry, enable quantitative analysis of physiological and pathological processes. For Ferucarbotran, coating and size distribution of the core had to be considered to match experimental data.</dc:description>

Bioabsorbable metals offer great promise for temporary medical implants. Among them, iron (Fe) and magne sium (Mg) alloys are of particular interest due to their biocompatibility, mechanical properties, and degradation behavior. However, Fe degrades too slowly, while Mg degrades too quickly. Combining both elements has been proposed to overcome these drawbacks, though their immiscibility presents a processing challenge. In this study, Fe-5 wt% Mg powder was produced via high-energy ball milling (Fe5Mg-BM), and its degradation was compared with pure Fe (Fe99) and milled Fe (Fe-BM). In vitro degradation was evaluated in three simulated body fluids:
phosphate-buffered saline (PBS), modified Hanks’ balanced salt solution (HBSS), and Dulbecco’s Modified Eagle Medium (DMEM). Ion release, pH changes, and degradation products were characterized, along with cytotoxicity and antibacterial activity. The results showed that Fe5Mg-BM exhibited accelerated degradation compared to Fe99 and Fe-BM. Among the media, modified HBSS induced the greatest diversity and quantity of degraded compounds. Ion analysis revealed negligible iron release in PBS and HBSS, with the highest levels in DMEM for Fe5Mg-BM. Magnesium release followed the trend: PBS < DMEM < HBSS. Fe5Mg-BM showed limited cytotox icity at 3 days and enhanced antibacterial activity in PBS and HBSS compared to Fe-BM. These results indicate that the biological effects arise from complex interactions at the material interface and the solubility of degra dation products, suggesting that Mg is a promising alloying element for enhancing the performance of Fe-based materials in biomedical applications.

Background/Objectives: Dravet Syndrome (DS) is a severe form of epilepsy that typically manifests in the first year of life and often requires polytherapy with two or more anti- seizure medications (ASMs) to achieve adequate seizure control. Whereas the combination of stiripentol (STP) and cannabidiol (CBD) has demonstrated clinical efficacy, it presents significant formulation challenges due to the low aqueous solubility and poor oral bioavail- ability of both compounds. Furthermore, the high daily dosages of STP (approximately 50 mg/kg/day or higher) and the oily nature of conventional CBD formulations often hinder patient compliance, as pediatric patients frequently reject these treatments due to unfavorable organoleptic properties. Methods: Nanostructured lipid carriers (NLCs) con- taining STP and CBD suspended in an aqueous medium were developed. The formulation was optimized using Response Surface Methodology (RSM) and subjected to comprehen- sive in vitro and in vivo characterization. Results: The optimized formulation exhibited a mean particle size of 175.3 nm, a polydispersity index (PDI) of 0.232, a zeta potential of −8.35 mV, and an encapsulation efficiency greater than 99% for both drugs. Physico- chemical characterization via atomic force microscopy, differential scanning calorimetry, thermogravimetric analysis, X-ray diffraction, and Fourier transform infrared spectroscopy revealed spherical nanoparticles without aggregation, with the drugs molecularly dis- persed within the lipid matrix. Both STP and CBD showed sustained release profiles and demonstrated oral pharmacokinetic profiles that were comparable or superior to current commercial products. Conclusions: This novel formulation represents a promising thera- peutic alternative for DS, enabling the co-administration of STP and CBD while potentially enhancing CBD bioavailability and treatment adherence in pediatric populations.

This work reports a detailed theoretical study of the molecular parameters, harmonic vibrational frequencies, UV absorption spectra and standard enthalpies of formation for the radicals C₂X₃ (with X = F, Cl and Br) and a comparison with the corresponding determinations for the rest of the members of the family C₂Xₙ (with n = 2-4). Molecular properties were calculated using different levels of theory:
density functional theory employing the B3LYP, X3LYP, BMK, M06-2X and M08-HX functionals combined with the basis sets 6–311++G(3df,3pd) and aug-cc-pVTZ, and the ab initio composite models G3B3 and G4. Structural and spectroscopic characterization of the C₂F₃, C₂Cl₃ and C₂Br₃ radicals, along with the estimation of the enthalpies of formation of C₂F₃ and C₂Cl₃, were derived here for the first time, to our knowledge. Accuracy in the quantification of ¹³¹ I activity from spect images: a SIMIND Monte Carlo study
Anales AFA; vol. 37, no. 1
The possibility of quantifying the activity of a radiopharmaceutical from SPECT images depends, among others, on the technical specifications of the imaging equipment, the isotope/collimator combination, and the presence of physical phenomena experienced by the emitted radiation that degrades the image. These factors can be investigated and optimized using Monte Carlo-based simulations. The aim of this work is to evaluate the accuracy on the quantification of ¹³¹ I activity from SPECT images obtained with a General Electric SPECT equipment, model Discovery NM 630, under different operational conditions. To this end, a gamma camera was configured in SIMIND MC as a computational model of the clinical equipment and lesions containing ¹³¹ I in different locations of an anthropomorphic phantom was configured as a model of the patient. Simulated SPECT images were obtained using general purpose medium- and high- energy collimators and different thicknesses of scintillation crystal, and ¹³¹ I activity was quantified from the images, with and without attenuation correction. The quantified ¹³¹ I activity from simulated SPECT images without attenuation correction is always smaller than the activity configured in the simulation, yielding errors of between 39 and 64 %. The error in the quantification of ¹³¹ I activity markedly decreases when the image is corrected for attenuation, yielding error values less than 0.4 % in the thyroid remnants. The work verified that it is possible to quantify ¹³¹ I activity from SPECT images if an attenuation correction method is available.

In this work, we evaluate the efficiency of a DNA purification protocol from gynecological samples using locally synthesized Fe3 O4 @SiO2 magnetic microparticles and a low-cost, guanidinium thiocyanate (GITC)-free lysis buffer. The microparticles were characterized by SEM, EDS, FTIR, and magnetic measurements, confirming the formation of compact silica- coated aggregates with suitable magnetic responsiveness for rapid and complete capture.
Using this material in combination with a simple, GITC-free lysis buffer, we achieved DNA extraction yields comparable to those obtained with standard methods based on chaotropic salts. The purified DNA showed high compatibility with molecular assays for the detection of Chlamydia trachomatis, Ureaplasma urealyticum, Mycoplasma hominis, and human papilloma virus. Clinical validation demonstrated excellent diagnostic performance, with only a few discrepancies observed in samples near the detection threshold of qPCR, a limitation shared with commercial kits. Overall, the method represents a low-cost, safe, and sustainable alternative for routine clinical and epidemiological applications, compared to methods based on chaotropic salt buffers. Furthermore, it reduces reliance on imported commercial consumables and minimizes the handling of hazardous reagents.

Sleep is known to support memory consolidation through a complex interplay of neural dynamics across multiple timescales. Using intracranial EEG (iEEG) recordings from patients undergoing clinical monitoring, we characterize spectral activity, neuronal avalanche dynamics, and temporal correlations across sleep-wake states, with a focus on their spatial distribution and potential functional relevance. We observe increased low-frequency power, larger avalanches, and enhanced long-range temporal correlations-quantified via Detrended Fluctuation Analysis-during N2 and N3 sleep. In contrast, REM sleep and wakefulness show reduced temporal persistence and fewer large-scale cascades, suggesting a shift toward more fragmented and flexible dynamics. These signatures vary across cortical regions, with distinctive patterns emerging in medial temporal and frontal areas-regions implicated in memory processing. Rather than providing direct evidence of consolidation, our results point to a functional neural landscape that may favor both stabilization and reconfiguration of internal representations during sleep. Overall, our findings highlight the utility of iEEG in revealing the multiscale spatio-temporal structure of sleep-related brain dynamics, offering insights into the physiological conditions that support memory-related processing.
