<title>Cellular and molecular basis of progesterone-induced neuroprotection</title>
<link>http://sedici.unlp.edu.ar:80/handle/10915/127232</link>
<description>Revision
Physiological Mini Reviews; vol. 11, no. 6
Progesterone exerts several effects in the central nervous system not directly involved in reproduction or sex behavior. Non-reproductive effects are better observed under pathological conditions, and include stimulation of myelin formation, neurogenesis and neurosteroidogenesis, preserved mitochondrial function, neuroprotection, anti-inflammatory effects, decreased glutamate excitotoxicity, and regulation of mood, memory and cognition. In addition, the progesterone reduced derivative allopregnanolone shows anxiolytic, sedative and anesthetic properties after binding to GABAa receptors. In the present report we provide examples of [1] progesterone effects on the local synthesis of steroids (“neurosteroids”) in a demyelination model, [2] the requirement of the classical progesterone receptor for the antiinflammatory effects in mice with spinal cord injury, and [3] the protective role of progesterone and allopregnanolone in a mouse model of neurodegeneration. In conclusion, the beneficial effects observed in different experimental paradigms support the versatile properties of progesterone in animal models of central nervous system disorders.; La progesterona produce varios efectos en el sistema nervioso central no relacionados a la reproducción o comportamiento sexual.

<title>Microtubules and mitochondria nanotunnels</title>
<link>http://sedici.unlp.edu.ar:80/handle/10915/127230</link>
<description>Revision
Physiological Mini Reviews; vol. 11, no. 5
Mitochondrial function strictly depends on mitochondrial structure, location and dynamics.&#13;
Mitochondria and cytoskeleton interaction is crucial for normal mitochondrial morphology distribution and motility. In cardiac muscle, microtubules sustain muscle contraction and mitochondria disposition; the latter is crucial for localized uptake of calcium (Ca2+) and consequentially energy production needed for cardiac function.&#13;
Few studies have been conducted on understanding mitochondria and microtubules interaction in cardiomyocytes. In cardiac muscle, recent studies have shown that cardiac mitochondria do not need to migrate to communicate with each other, but they can extend protrusions called nanotunnels to reach and communicate with other mitochondria in long-distance. This process does not involve mitochondria movement, but possibly involves a pulling action along microtubules. The association of mitochondria nanotunnels and microtubules have been identified using 2D and 3D electron microscopy [1]. These new findings highlight the need to investigate the action of microtubules on cardiac mitochondria and its effect on mitochondria dynamics and cardiac function.

<title>The immediately releasable pool of secretory vesicles in adrenal chromaffin cells</title>
<link>http://sedici.unlp.edu.ar:80/handle/10915/127228</link>
<description>Revision
Physiological Mini Reviews; vol. 11, no. 4
The exocytosis of hormone-filled vesicles in adrenal chromaffin cells is triggered by localized Ca²⁺ gradients that develops after the activation of voltage-dependent Ca²⁺  channels. To reach the fusion competent state the vesicles go through a variety of maturation steps, including the mobilization through the cytoskeleton, the docking to membrane, and the priming of the exocytotic molecular machinery. However, the fusion readiness of vesicles will also depend on their proximity to the Ca²⁺  source. The immediately releasable pool is a small group of ready–to-fuse vesicles, whose fusion is tightly coupled to Ca²⁺  entry through channels. Recent work indicates that such a coupling is not produced by a random distribution between parts, but is the result of a specific interaction of secretory vesicles with P/Q-type Ca²⁺  channels. The immediately releasable pool is able to sustain with high efficiency the secretion triggered by brief depolarizations applied at low frequencies, like action potentials at basal conditions in adrenal chromaffin cells.; La exocitosis de las vesículas secretorias en células cromafines adrenales es disparada por gradientes de Ca²⁺  localizados que se desarrollan a partir de la activación de canales de Ca²⁺  voltaje dependiente. Para poder fusionarse a la membrana, las vesículas deben pasar primero por una serie de etapas madurativas que incluyen el transporte por el citoesqueleto, el anclado a la membrana y el priming de la maquinaria exocítica. Sin embargo, la eficiencia con que la vesícula se fusiona depende también de su proximidad con la fuente de Ca²⁺ calcio.&#13;

<title>Ryanodine receptor mutations: in sickness and in health</title>
<link>http://sedici.unlp.edu.ar:80/handle/10915/127226</link>
<description>Revision
Physiological Mini Reviews; vol. 11, no. 3
Ryanodine Receptors (RyR) are large ion channels necessary for Ca2+ release from intracellular stores in many cell types. Since RyR was discovered nearly 30 years ago, a fruitful research field has devoted to understanding the physiological regulation of the three protein isoforms in health, and the mechanisms underlying their dysfunction in sickness. This minireview discusses the structural characteristics of RyR2, the cardiac isoform, its regulation during cardiac function and the clinical significance of specific mutations. While hundreds of RyR2 mutations are associated with cardiac disease with different levels of confidence, close to 1500 variants appear in the general population without inducing harmful phenotypes. Hence, studying RyR2 mutations variants may shed light on overall protein regulation and the mechanisms that compensate for constitutive changes in RyR2 function.

<title>More than tracks: how the cytoskeleton builds epithelia asymmetry</title>

<description>Revision
Physiological Mini Reviews; vol. 11, no. 2
Epithelia are three-dimensional arrangements of cells organized into structures which delimit morphologically and physiologically different compartments. At the cellular level, epithelial cells are characterized by the presence of an apical pole, facing the lumen of an organ or the outer environment, and baso-lateral domains, involved in cell-extracellular matrix and cell-cell interactions. Each pole is characterized by its specific protein and lipid membrane composition, and its distinct assembly of cellular organelles. This polarized organization of cell components ensures the vectorial absorption and secretion of proteins and other molecules, which constitute the essential function of epithelia. Actin and microtubule cytoskeletons are also polarized in epithelial cells. This organization contributes to the de novo establishment as well as to the functional maintenance of epithelial polarity. In this article we delineate the central features of epithelial polarity and summarize the current knowledge on how actin and microtubule cytoskeletons contribute to epithelial organization.; Los epitelios son arreglos tridimensionales de células, organizados en estructuras que delimitan compartimientos morfológica y fisiológicamente diferentes.

<title>Nitroso-redox balance: a key mechanism in the regulation of the myocardial function in health and disease</title>
<link>http://sedici.unlp.edu.ar:80/handle/10915/127221</link>
<description>Revision
Physiological Mini Reviews; vol. 11, no. 1
Given the close interaction between nitric oxide (NO) and reactive oxygen species (ROS) in biological systems and their especial relevance in regulating aspects of the cardiovascular physiology, the concept of “nitroso-redox balance” has arisen as a more comprehensive manner of interpreting the intracellular redox state. Nitroso-redox signaling pathways participate in numerous cardiovascular mechanisms, including myocardial contractility and relaxation, mitochondrial respiration, and endothelial function. Alterations of this balance are involved in numerous aspects of cardiovascular pathophysiology. NO and ROS generating mechanisms play a major role in both regulating and responding to the redox state of the cell, which targets calcium handling, contractile and vasoactive mechanisms. Thus, the nitroso-redox signaling pathway is critically important in cardiac physiology and pathophysiology, and consequently a fundamental therapeutic target.
