The long-term storage of citrus germplasm requires analyzing the desiccation tolerance of whole seeds. Citrus limon L. Burm vc. eureka seeds were desiccated using 8 relative humidities (HR%) until equilibrium conditions were reached (range of 11-95% at 20 ºC) and the percentage of germination (% G) was determined for each case. The chemical composition of seeds and water activity were obtained. The moisture sorption isotherm was determined and modeled using the GAB and Darcy & Watt equations and the water content as a function of the desiccation period was evaluated. Differential scanning calorimetry (DSC) was applied to identify the water and lipid melting transitions. The seed presented an initial humidity of 44.67% (w.b.) and a lipid content of 22.21% (w.b.). The GAB monolayer water content was 0.080 (g water / g dry basis); the Darcy & Watt model shows a better fitting of experimental results. The %G was quantified at 9, 16, 21 and 25 days and significant differences were observed between treatments with different HR%. Citrus seeds that were partially desiccated in the range of 11 a 33% HR shows significant differences with seeds without treatment having %G between 80-83.3%.
